Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Peptide degradation follows several routes. Hydrolysis cleaves the backbone at susceptible residues, oxidation targets methionine and tryptophan side chains, and aggregation produces higher-molecular-weight species that are difficult to reverse. Light exposure accelerates oxidation, which is why amber glass or opaque secondary packaging is common. Repeated freeze-thaw cycles promote aggregation and are best avoided. Stability-indicating methods detect these changes before they become visible.
Quality control for research material typically involves reversed-phase HPLC for purity and identity, mass spectrometry for molecular weight confirmation, and Karl Fischer titration for residual water content. Peptide content is often reported as the mass of actual peptide rather than total powder mass, since counterions and water contribute to the latter. A certificate of analysis should list the method used for each specification. Limits and acceptance criteria vary by supplier and by intended application.

As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
=== Safety === Mild to moderate gastrointestinal side effects (such as nausea, cramping, bloating, and abdominal discomfort) are common after ESG (reported in over 70% of patients), but these are predictable, temporary, and can be managed with medications. Most will resolve within one week after ESG. Due to the minimally invasive nature of ESG, serious complications are rare. These include pain or nausea requiring hospitalization (1.08%); upper gastrointestinal tract bleeding (0.56%); peri-gastric leak or infected fluid collection (0.48%); pulmonary embolism (0.06%); perforation (0.06%). Similar rates of serious adverse events were reported in the multicenter, randomized controlled MERIT study. No deaths have been observed in the published ESG literature.
=== Denaturing-condition electrophoresis === Gel electrophoresis is a common laboratory technique that can be used both as a preparative and analytical method. The principle of electrophoresis relies on the movement of a charged ion in an electric field. In practice, the proteins are denatured in a solution containing a detergent (SDS). In these conditions, the proteins are unfolded and coated with negatively charged detergent molecules. The proteins in SDS-PAGE are separated on the sole basis of their size. In analytical methods, the protein migrate as bands based on size. Each band can be detected using stains such as Coomassie blue dye or silver stain. Preparative methods to purify large amounts of protein, require the extraction of the protein from the electrophoretic gel. This extraction may involve excision of the gel containing a band, or eluting the band directly off the gel as it runs off the end of the gel. In the context of a purification strategy, denaturing condition electrophoresis provides an improved resolution over size exclusion chromatography, but does not scale to large quantity of proteins in a sample as well as the late chromatography columns.
The NRD pathway for the 40S subunit may be independent or separate from the NRD pathway for the 60S subunit. It has been observed that certain genes were able to affect degradation of certain pre-RNAs, but not others. Numerous proteins are involved in the NRD pathway, such as Mms1p and Rtt101p, which are believed to complex together to target ribosomes for degradation. Mms1p and Rtt101p are found to bind together and Rtt101p is believed to recruit a ubiquitin E3 ligase complex, allowing for the non-functional ribosomes to be ubiquinated before being degraded. Prokaryotes lack a homolog for Mms1, so it is unclear how prokaryotes are able to degrade non-functional rRNAs. The growth rate of eukaryotic cells did not seem to be significantly affected by the accumulation of non-functional rRNAs.

=== Prognosis === In general, the presence and density of cancer associated fibroblasts (CAF) point towards a bad prognosis for the patient, and so, are pro-tumour. These could however be used as markers for diagnosis and therapies, thus diagnosing at an earlier stage. The presence of podoplanin in CAFs has been found to play a fundamental role in worsening the prognosis of patients with lung adenocarcinoma; this could however be helpful as a marker to diagnose at an early stage. In oesophageal adenocarcinomas, CAFs release the ECM (Extracellular Matrix) protein periostin and promote tumour cell growth through paracrine signalling. However, blocking specific integrin receptors and pathways can ceases the invasion of tumor cells. The greater the density of CAFs found in oral cancer, the poorer the prognosis, as this significantly decreases the 5 year survival rate. Being female in this study also proved to be a bigger risk factor, with men being protected more against the effects.
== Origins of the term == The term AI Cold War first appeared in 2018 in an article in Wired magazine by Nicholas Thompson and Ian Bremmer. The two authors trace the emergence of the AI Cold War narrative to 2017, when China published its AI Development Plan, which included a strategy aimed at becoming the global leader in AI by 2030. While the authors acknowledge the use of AI by China to strengthen its authoritarian (totalitarian) rule, they warn against the perils for the US of engaging in an AI Cold War strategy. Thompson and Bremmer rather advocate for a technological cooperation between the US and China to encourage global standards in privacy and ethical use of AI. Shortly after the publication of the article in Wired magazine, the former U.S. Treasury Secretary Hank Paulson referred to the emergence of an ‘Economic Iron Curtain’ between the US and China, reinforcing the new AI Cold War narrative.
== Taxonomy == Hybridization studies from 1995 indicate that species within the genus Micrococcus are not closely related, showing as little as 50% sequence similarity. This suggests that some Micrococcus species may, on the basis of ribosomal RNA analysis, eventually be re-classified into other microbial genera.

The ELN denied involvement with the alleged drug boat or any other drug boat trafficking. On 1 November, a strike "at the direction of President Trump" according to Hegseth, killing three people took place on what the US claimed was a designated terrorist organization, though the organization's name was not given. Hours after the US Senate voted down a resolution that would have required congressional approval for further strikes, a 6 November strike in the Caribbean killed three people, with debris and body parts reportedly washing ashore in Colombia days later. A strike on 10 November in the Caribbean killed four people. A 13 February strike killed three people. Three strikes were conducted on 16 February, one of which killed three people in the Caribbean. A 23 February strike killed three people.A 25 March strike killed four people.
=== Early work === Thomson's prize-winning master's work, Treatise on the motion of vortex rings, shows his early interest in atomic structure. In it, Thomson mathematically described the motions of Lord Kelvin's vortex theory of the atom. Thomson published a number of papers addressing both mathematical and experimental issues of electromagnetism. He examined the electromagnetic theory of light of James Clerk Maxwell, introduced the concept of electromagnetic mass of a charged particle, and demonstrated that a moving charged body would apparently increase in mass. Much of his work in mathematical modelling of chemical processes can be thought of as early computational chemistry. In further work, published in book form as Applications of dynamics to physics and chemistry (1888), Thomson addressed the transformation of energy in mathematical and theoretical terms, suggesting that all energy might be kinetic. His next book, Notes on recent researches in electricity and magnetism (1893), built upon Maxwell's Treatise upon electricity and magnetism, and was sometimes referred to as "the third volume of Maxwell." In it, Thomson emphasized physical methods and experimentation and included extensive figures and diagrams of apparatus, including a number for the passage of electricity through gases. His third book, Elements of the mathematical theory of electricity and magnetism (1895) was a readable introduction to a wide variety of subjects, and achieved considerable popularity as a textbook.
== Pull down time == The pull down time is defined as the necessary time to cool down the ULT freezer from ambient temperatures to the selected temperature of −80 to −86 °C (−112 to −123 °F). The time strongly depends on the type of insulation, the efficiency of the compressor system as well as the installed metal shelves within the freezer. At the start of the twenty-first century, ULT freezers were able to cool down within 3 to 5 hours. Warm up time is typically 1/8 °C per minute.

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
Structural modifications, including a fatty acid side chain and non-natural amino acid substitutions, slow enzymatic breakdown and promote albumin binding. These changes support once-weekly dosing rather than twice-daily administration.
The pathways involving insulin, glucagon, gastric emptying, and appetite signaling are well described. How much each pathway contributes to weight reduction in a given person is not fully established.
