The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.

Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
== Legacy == Harald Pedersen's engineering contributions helped developing the early Danish insulin industry. He and his brother also established the Novo Foundation in 1951, of which Harald became a life-member of the Board. The Pedersen brothers founded Novo Terapeutisk Laboratorium in 1925 and competed with Nordisk until the 1989 merger that created Novo Nordisk.
=== Pharmacokinetics === Detailed pharmacokinetic studies on arsenic trioxide have not been conducted. When administered intravenously, a steady state is reached after 8–10 days. Arsenic binds to proteins to an insignificant extent. The highest concentrations of arsenic are found in the liver, kidneys, heart, lungs, hair, and nails. Arsenous acid is oxidized to arsenic acid and methylated in the liver, and then excreted 60% in the urine. The drug has a half-life of 92 hours. Arsenic trioxide is neither a substrate nor an inhibitor of cytochrome P450 isozymes (1A2, 2A6, 2B6, 2C8, 2C9, 2C19, 2D6, 2E1, 3A4/5, 4A9/11).
=== Dosages === For prostate cancer, the dosage of buserelin by subcutaneous injection is 500 μg three times per day (once every 8 hours, 1,500 μg/day total) for one week and then 200 μg once daily thereafter. If buserelin is used as a nasal spray, the dosage for prostate cancer is 800 μg sprayed into the nostrils three times per day (once every 8 hours, 2,400 μg/day total) for one week followed by 400 μg sprayed into the nostrils three times per day (once every 8 hours, 1,200 μg/day total) thereafter. For endometriosis, buserelin is used specifically as a nasal spray and the dosage is the same as that used for prostate cancer. These dosages of buserelin for both subcutaneous injection and nasal spray have been found to decrease testosterone levels to near-castrate levels in men with prostate cancer, although suppression was more complete with subcutaneous injection presumably due to suboptimal absorption with intranasal administration.
In a nuclear reactor, such a chain reaction is slowed and controlled by a neutron poison, absorbing some of the free neutrons. Such neutron-absorbent materials are often part of reactor control rods (see nuclear reactor physics for a description of this process of reactor control). Other naturally occurring isotopes such as uranium-238 are fissionable, but not fissile, meaning that they only undergo fission when absorbing high energy (fast) neutrons. As little as 15 lb (6.8 kg) of uranium-235 can be used to make an atomic bomb. The nuclear weapon detonated over Hiroshima, Japan, named Little Boy, relied on uranium fission. However, the first nuclear bomb (the Gadget used at Trinity) and the bomb that was detonated over Nagasaki, Japan (Fat Man), were both plutonium bombs. Uranium metal has three allotropic forms:
As a result of canonical DNA sequence changes, directed evolution also enables the development of orthogonal ribosomes, which make it easier to incorporate XAAs into proteins or create "mirror life," or biological systems that contain biomolecules made up of enantiomers with different chiral orientations.

Chilling revelations of torture, rape, massacres, extermination were revealed through the 2014 Caesar Report, which documented photographic evidences of industrial-scale atrocities occurring in Syrian military prisons. The report documented a total of 55,000 digital images of tortured or dismembered human bodies of around 11,000 detainees. Describing some of the torture techniques unleashed on Syrians held captive in military prisons, the military defector Caesar states: "It was very clear that they were tortured, not tortured for a day or two, tortured for many, many long months. They were emaciated bodies, purely skeletons. There were people, most of them had their eyes gouged out. There was electrocution, you could tell by the dark spots on their body that was used there. There was utilization of knives and also big cables and belts that was used to beat them. And so, we could see every type of torture on the bodies of these individuals. 'Every type of torture,' but the depravity of the gouged eyes leaves to the imagination how maiming was calculated to coerce information. By 2013, the bodies overflowed the morgues and spilled across a parking garage at a military hospital."In 2023, Canada and Netherlands jointly filed a lawsuit against the Assad regime at the International Court of Justice (ICJ); charging Assad with ordering torture, rapes and other de-humanising tactics on hundreds of thousands of detainees in Syrian prison networks, including women and children.
==== Thin-client ==== A thin-client LIMS is a more modern architecture which offers full application functionality accessed through a device's web browser. The actual LIMS software resides on a server (host) which feeds and processes information without saving it to the user's hard disk. Any necessary changes, upgrades, and other modifications are handled by the entity hosting the server-side LIMS software, meaning all end-users see all changes made. To this end, a true thin-client LIMS will leave no "footprint" on the client's computer, and only the integrity of the web browser need be maintained by the user. The advantages of this system include significantly lower cost of ownership and fewer network and client-side maintenance expenses. However, this architecture has the disadvantage of requiring real-time server access, a need for increased network throughput, and slightly less functionality. A sort of hybrid architecture that incorporates the features of thin-client browser usage with a thick client installation exists in the form of a web-based LIMS. Some LIMS vendors are beginning to rent hosted, thin-client solutions as "software as a service" (SaaS). These solutions tend to be less configurable than on-premises solutions and are therefore considered for less demanding implementations such as laboratories with few users and limited sample processing volumes. Another implementation of the thin client architecture is the maintenance, warranty, and support (MSW) agreement.
Treatment is mainly supportive (gastric lavage, activated carbon, fluid resuscitation) but includes various drugs to counter the amatoxins, including intravenous penicillin and cephalosporin derivatives, and, in cases of greater ingestion, can extend to an orthotopic liver transplant. The most reliable method to treat amanitin poisoning is through gastric lavage immediately after ingestion; however, the onset of symptoms is generally too late for this to be an option. Chemically modified silibinin, silibinin dihydrogen disuccinate disodium (trade name Legalon SIL) a solution for IV administration, is used in treatment of severe intoxications with hepatotoxic substances such as paracetamol and amanitins.
=== Benzodiazepines === Benzodiazepines are used for acute anxiety and could be added along with current use of SSRIs to stabilize a treatment. Long-term use in treatment plans is not recommended. Different kinds of benzodiazepine will vary in its pharmacological profile, including its strength of effect and time taken for metabolism. The choice of the benzodiazepine will depend on the corresponding profiles. Benzodiazepines are used for emergent or short-term management. They are not recommended as the first-line anti-anxiety drugs, but they can be used in combination with SSRIs/SNRIs during the initial treatment stage. Indications include panic disorder, sleep disorders, seizures, acute behavioral disturbance, muscle spasm and premedication and sedation for procedures.
Biden was elected the 46th president in November 2020, defeating the incumbent, Donald Trump. Trump and numerous other Republicans repeatedly made false claims that widespread electoral fraud had occurred and that only he had legitimately won the election. Biden's transition was delayed by several weeks as the White House ordered federal agencies not to cooperate. On November 23, General Services Administrator Emily W. Murphy formally recognized Biden as the apparent winner of the 2020 election and authorized the start of a transition process to the Biden administration. Although most resulting lawsuits were either dismissed or ruled against by numerous courts, Trump nonetheless conspired with his campaign team to submit documents in several states (all of which Biden had won) that falsely claimed to be legitimate electoral votes for President Trump and Vice President Mike Pence. After the submission of these documents, the Trump campaign intended that the presiding officer of the United States Senate, either President of the Senate Pence or President pro tempore Chuck Grassley, would claim the unilateral power to reject electors during the January 6, 2021, vote counting session; the presiding officer would reject all electors from the several states for which the Trump campaign had submitted false documents, leaving 232 votes for Trump and 222 votes for Biden, thereby overturning the election results in Trump's favor. This plan failed after Pence refused to cooperate with it.

Neuroleptic, originating from the Ancient Greek terms νεῦρον ('neuron') and λαμβάνω ('to take hold of')—thus meaning 'which takes the nerve'—refers both to common neurological effects and to side-effects. An estimated 1.7% of adults in the United States take antipsychotics, according to a 2018 study.
At least three isoforms of synuclein are produced through alternative splicing. The majority form of the protein, and the one most investigated, is the full-length protein of 140 amino acids. Other isoforms are alpha-synuclein-126, which lacks residues 41-54 due to loss of exon 3; and alpha-synuclein-112, which lacks residues 103-130 due to loss of exon 5.
During this period, royalist forces made advances into New Granada, which they controlled from 1815 to 1819, and into Chile, which they controlled from 1814 to 1817. Except for royalist areas in the northeast and south, the provinces of New Granada had maintained independence from Spain since 1810, unlike neighboring Venezuela, where royalists and pro-independence forces had exchanged control of the region several times. To pacify Venezuela and to retake New Granada, Spain organized in 1815 the largest armed force it ever sent to the New World, consisting of 10,500 troops and nearly sixty ships. Although this force was crucial in retaking a solidly pro-independence region like New Granada (see Spanish reconquest of New Granada), its soldiers were eventually spread out throughout Venezuela, New Granada, Quito, and Peru, and were lost to tropical diseases, diluting their impact on the war. More importantly, the majority of the royalist forces were composed, not of soldiers sent from the peninsula, but of Spanish Americans. Overall, Europeans formed only about a tenth of the royalist armies in Spanish America, and only about half of the expeditionary units, once they were deployed in the Americas. Since each European soldier casualty was replaced by a Spanish American soldier, over time, there were more and more Spanish American soldiers in the expeditionary units.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
The lyophilized powder is dissolved in a suitable solvent, often sterile water or a buffered diluent, with gentle mixing rather than vigorous shaking. Foaming and shear should be avoided because they can promote aggregation. The resulting solution is then stored cold and protected from light.
