Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.

Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
== Discovery == The presence of the nuclear membrane that sequesters the cellular DNA is the defining feature of eukaryotic cells. The nuclear membrane, therefore, separates the nuclear processes of DNA replication and RNA transcription from the cytoplasmic process of protein production. Proteins required in the nucleus must be directed there by some mechanism. The first direct experimental examination of the ability of nuclear proteins to accumulate in the nucleus was carried out by John Gurdon when he showed that purified nuclear proteins accumulate in the nucleus of frog (Xenopus) oocytes after being micro-injected into the cytoplasm. These experiments were part of a series that subsequently led to studies of nuclear reprogramming, directly relevant to stem cell research. The presence of several million pore complexes in the oocyte nuclear membrane and the fact that they appeared to admit many different molecules (insulin, bovine serum albumin, gold nanoparticles) led to the view that the pores are open channels and nuclear proteins freely enter the nucleus through the pore and must accumulate by binding to DNA or some other nuclear component. In other words, there was thought to be no specific transport mechanism. This view was shown to be incorrect by Dingwall and Laskey in 1982. Using a protein called nucleoplasmin, the archetypal ‘molecular chaperone’, they identified a domain in the protein that acts as a signal for nuclear entry.
=== Interest in foreign technology === The Soviets tried to import technology to solve the issues with the powerplant, avionics and manufacturing capacity. The attempts took two directions. First was wholesale technology transfer similar to the Li-2 deal of the 1930s. This would have delayed Il-86 development, since the programme would have been demoted to a reserve status. The second direction was to import individual systems and items. This would have speeded Il-86 development. The fact that Il-86 development was protracted indicates that for long periods the programme was pursued as backup insurance in case wholesale technology transfer failed. Before the Boeing 747 had flown, a Ministry of Civil Aviation delegation visited the United States for a series of detailed sales presentations on the type lasting three days. At the 1971 Paris Salon, Ilyushin bureau head Genrikh Novozhilov and Boeing's Joe Sutter are claimed to have arranged an informal technology trade-off. Over supper in a Paris restaurant, the Soviet side ceded information on titanium technology to the Americans, while the latter, "sketching on the tablecloth," ceded information on pylon-mounted podded engines and "the structural and aerodynamic amity of the aeroelastic wing." Soviet interest in buying the 747s continued until the end of détente in the late 1970s. At the peak of détente, on March 11, 1974, a Lockheed L-1011 TriStar arrived in Moscow for three days of sales presentations and demonstrations. The TriStar matched the Il-86 in size and performance and had development potential.
Money, or its hectic pursuit, has been shown to hinder people's savoring ability, or the act of enjoying everyday positive experiences and emotions. In a study looking at working adults, wealthy individuals reported lower levels of savoring ability (the ability to prolong positive emotion) relative to their poorer peers. Studies have routinely shown that nations are happier when people's needs are met. Some studies suggest, however, that people are happier after spending money on experiences, rather than physical things, and after spending money on others, rather than themselves. However, purchases that buy 'time', for instance, cleaners or cooks typically increase individual well-being. Lottery winners report higher levels of happiness immediately following the event. But research shows winner's happiness levels drop and return to normal baseline rates within months to years. This finding suggests money does not cause long-term happiness (1978). However, in a more recent British study on lottery prizes between £1,000 and £120,000, a positive effect even two years after the event was found, the return to normal being only partial and varying. One 600 women strong 2011 study shows that house owners are no happier than renters. Degree of ownership also matter: "...housing property rights matter for subjective well-being.

The game's production was turbulent, as the design's scope exceeded the available resources, and the team was left without a producer for nearly a year until Activision appointed David Mullich to the role, where he found designs and levels unfinished or abandoned. After three years in development with no end in sight and running over budget, Activision set a strict deadline for completion, and Bloodlines was released incomplete in November 2004. Released in competition with Half-Life 2 and several other titles, Bloodlines sold fewer than 80,000 copies during its initial release, which was considered a poor performance. It received generally positive reviews from contemporary critics, who praised the game's writing and the scale of choice, although they criticized its technical flaws. It was Troika Games's last production before its failure in early 2005, when it could not secure additional projects. The game has a cult following as a rarely replicated example of gameplay and narrative, and modern reception recognizes it as a flawed masterpiece. Since its original release, Bloodlines received post-release support from fans, supplying unofficial fixes and re-adding unused content. A sequel, Vampire: The Masquerade – Bloodlines 2, was released in 2025.
== Reception == The game was generally well received. Charlie Hill of Polygon called it "absolutely brilliant". Aaron Zimmerman stated in a review for Ars Technica that the artwork is "top-notch" and the card stock is of good quality.
=== London Fire Brigade === Brent has three fire stations: Park Royal, Wembley and Willesden. Brent has a mixture of residential, industrial and commercial land. Wembley National Stadium is in the borough; on match days the fire safety of over 90,000 people falls to the London Fire Brigade. The Wembley station covers the largest area in the borough, 19.1 km2 (7.4 sq mi). Two pumping appliances, a fire rescue unit and an aerial ladder platform are based there. Willesden, for its more typical area covered (10.5 km2 (4.1 sq mi)), responded to over a thousand incidents in 2006/2007. Two pumping appliances reside there. Park Royal, with its one pumping appliance and an incident response unit covers 8.1 km2 (3.1 sq mi). Within the borough, 4,105 incidents occurred in 2006/2007.
Infrared spectroscopy exploits the fact that molecules absorb frequencies that are characteristic of their structure. These absorptions occur at resonant frequencies, i.e. the frequency of the absorbed radiation matches the vibrational frequency. The energies are affected by the shape of the molecular potential energy surfaces, the masses of the atoms, and the associated vibronic coupling.

=== Genomics and bioinformatics tools to study browning === Due to the complex nature of adipose tissue and a growing list of browning regulatory molecules, great potential exists for the use of bioinformatics tools to improve study within this field. Studies of WAT browning have greatly benefited from advances in these techniques, as beige fat is rapidly gaining popularity as a therapeutic target for the treatment of obesity and diabetes. DNA microarray is a bioinformatics tool used to quantify expression levels of various genes simultaneously, and has been used extensively in the study of adipose tissue. One such study used microarray analysis in conjunction with Ingenuity IPA software to look at changes in WAT and BAT gene expression when mice were exposed to temperatures of 28 and 6 °C. The most significantly up- and downregulated genes were then identified and used for analysis of differentially expressed pathways. It was discovered that many of the pathways upregulated in WAT after cold exposure are also highly expressed in BAT, such as oxidative phosphorylation, fatty acid metabolism, and pyruvate metabolism. This suggests that some of the adipocytes switched to a beige phenotype at 6 °C. Mössenböck et al. also used microarray analysis to demonstrate that insulin deficiency inhibits the differentiation of beige adipocytes but does not disturb their capacity for browning. These two studies demonstrate the potential for the use of microarray in the study of WAT browning.
==== Comorbid personality disorders ==== Approximately 74% of individuals with BPD also fulfill criteria for another personality disorder during their lifetime, according to research published in 2008. The most prevalent co-occurring disorders are from Cluster A (paranoid, schizoid, and schizotypal personality disorders), affecting about half of those with BPD, with schizotypal personality disorder alone impacting one-third of individuals. Being part of Cluster B, BPD patients also commonly share characteristics with other Cluster B disorders (antisocial, histrionic, and narcissistic personality disorders), with nearly half of individuals with BPD showing signs of these conditions, and narcissistic personality disorder affecting roughly one-third. Cluster C disorders (avoidant, dependent, and obsessive-compulsive personality disorders) have the least comorbidity with BPD, with just under a third of individuals with BPD meeting the criteria for a Cluster C disorder.
Whereas in the course of human affairs history has shown that it may become necessary for a people to resolve the political affiliations which have connected them with another people and to assume amongst other nations the separate and equal status to which they are entitled: And Whereas in such event a respect for the opinions of mankind requires them to declare to other nations the causes which impel them to assume full responsibility for their own affairs: Now Therefore, We, The Government of Rhodesia, Do Hereby Declare: That it is an indisputable and accepted historic fact that since 1923 the Government of Rhodesia have exercised the powers of self-government and have been responsible for the progress, development and welfare of their people; That the people of Rhodesia having demonstrated their loyalty to the Crown and to their kith and kin in the United Kingdom and elsewhere through two world wars, and having been prepared to shed their blood and give of their substance in what they believed to be the mutual interests of freedom-loving people, now see all that they have cherished about to be shattered on the rocks of expediency; That the people of Rhodesia have witnessed a process which is destructive of those very precepts upon which civilization in a primitive country has been built, they have seen the principles of Western democracy, responsible government and moral standards crumble elsewhere, nevertheless they have remained steadfast; That the people of Rhodesia fully support the requests of their government for sovereign independence but have witnessed the consistent refusal of the Government of the United Kingdom to accede to their entreaties; That the Government of the United Kingdom have thus demonstrated that they are not prepared to grant sovereign independence to Rhodesia on terms acceptable to the people of Rhodesia, thereby persisting in maintaining an unwarrantable jurisdiction over Rhodesia, obstructing laws and treaties with other states and the conduct of affairs with other nations and refusing assent to laws necessary for the public good, all this to the detriment of the future peace, prosperity and good government of Rhodesia; That the Government of Rhodesia have for a long period patiently and in good faith negotiated with the Government of the United Kingdom for the removal of the remaining limitations placed upon them and for the grant of sovereign independence; That in the belief that procrastination and delay strike at and injure the very life of the nation, the Government of Rhodesia consider it essential that Rhodesia should attain, without delay, sovereign independence, the justice of which is beyond question; Now Therefore, We The Government of Rhodesia, in humble submission to Almighty God who controls the destinies of nations, conscious that the people of Rhodesia have always shown unswerving loyalty and devotion to Her Majesty the Queen and earnestly praying that we and the people of Rhodesia will not be hindered in our determination to continue exercising our undoubted right to demonstrate the same loyalty and devotion, and seeking to promote the common good so that the dignity and freedom of all men may be assured, Do, By This Proclamation, adopt, enact and give to the people of Rhodesia the Constitution annexed hereto;

Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
Albumin is the most abundant protein in plasma and carries molecules that bear fatty-acid chains. Binding shields the peptide from renal filtration and from peptidases, keeping a circulating reservoir. Slow release from this reservoir produces sustained receptor occupancy and supports infrequent dosing.
The insulinotropic effect is glucose-dependent, meaning secretion increases mainly when glucose is elevated. This property is often described as lowering the chance of hypoglycaemia when the compound is used alone. Other glucose-lowering agents used at the same time can still cause low blood glucose.
It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
