Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.

Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
CPT-symmetry puts strong constraints on the relative properties of particles and antiparticles and, therefore, is open to stringent tests. For example, the charges of a proton and antiproton must sum to exactly zero. This equality has been tested to one part in 108. The equality of their masses has also been tested to better than one part in 108. By holding antiprotons in a Penning trap, the equality of the charge-to-mass ratio of protons and antiprotons has been tested to one part in 6×109. The measured magnetic moments of protons and antiprotons have been found to be equal and opposite within 0.8 ppm.
It is possible Rose's increasing family size, plus the fact she and her husband had, by the mid-1970s, begun seeking avenues to exploit girls from care homes in addition to acquiring contacts—willing or unwilling—to submit to their fetishes, may have led the couple to decide that these avenues of control and domination were sufficient for their satisfaction.
==== Increased bone turnover ==== Retinoic acid suppresses osteoblast activity and stimulates osteoclast formation in vitro, resulting in increased bone resorption and decreased bone formation. It is likely to exert this effect by binding to specific nuclear receptors (members of the retinoic acid receptor or retinoid X receptor nuclear transcription family) which are found in every cell (including osteoblasts and osteoclasts). This change in bone turnover is likely to be the reason for numerous effects seen in hypervitaminosis A, such as hypercalcemia and numerous bone changes such as bone loss that potentially leads to osteoporosis, spontaneous bone fractures, altered skeletal development in children, skeletal pain, radiographic changes, and bone lesions.
antibody Any of a diverse family of glycoproteins known as immunoglobulins capable of binding specifically but reversibly via non-covalent interactions to a particular antigen or immunogen. Antibodies are generated as part of an organism's immune response to the introduction of a specific antigen into a host organism, and their binding of the antigen frequently (though not always) counteracts or inhibits any biological activity the antigen may have. Antibodies have a characteristic Y-shaped structure consisting of a heavy chain and light chain held together by disulfide bonds.
The Chartists gathered significant numbers around the People's Charter of 1838 which demanded the extension of suffrage to all male adults. Leaders in the movement also called for a more equitable distribution of income and better living conditions for the working classes. The very first trade unions and consumers' cooperative societies also emerged in the hinterland of the Chartist movement as a way of bolstering the fight for these demands. The Chartists were part of a Europe-wide wave of agitation for social reform and democratic rule, which peaked in the Revolutions of 1848. In France, the voice of this movement was La Montagne, also known as the Democratic Socialists. Karl Marx disliked La Montagne, viewing it as a party dominated by the middle class; he called them Sozialdemokrat, the first recorded use of the term social democracy. Around the same time, the British political philosopher John Stuart Mill also came to advocate a form of economic socialism within a liberal context known as liberal socialism. In later editions of Principles of Political Economy (1848), Mill would argue that "as far as economic theory was concerned, there is nothing in principle in economic theory that precludes an economic order based on socialist policies."

=== Biochemical tests === Fast and relatively simple biochemical tests can be used to identify infectious agents. For bacterial identification, the use of metabolic or enzymatic characteristics are common due to their ability to ferment carbohydrates in patterns characteristic of their genus and species. Acids, alcohols and gases are usually detected in these tests when bacteria are grown in selective liquid or solid media, as mentioned above. In order to perform these tests en masse, automated machines are used. These machines perform multiple biochemical tests simultaneously, using cards with several wells containing different dehydrated chemicals. The microbe of interest will react with each chemical in a specific way, aiding in its identification. Serological methods are highly sensitive, specific and often extremely rapid laboratory tests used to identify different types of microorganisms. The tests are based upon the ability of an antibody to bind specifically to an antigen. The antigen (usually a protein or carbohydrate made by an infectious agent) is bound by the antibody, allowing this type of test to be used for organisms other than bacteria. This binding then sets off a chain of events that can be easily and definitively observed, depending on the test. More complex serological techniques are known as immunoassays. Using a similar basis as described above, immunoassays can detect or measure antigens from either infectious agents or the proteins generated by an infected host in response to the infection.
== History == Esketamine was introduced for medical use as an anesthetic in Germany in 1997, and was subsequently marketed in other countries. In addition to its anesthetic effects, the medication showed properties of being a rapid-acting antidepressant, and was subsequently investigated for use as such. Esketamine received a breakthrough designation from the FDATooltip Food and Drug Administration for treatment-resistant depression (TRD) in 2013 and major depressive disorder (MDD) with accompanying suicidal ideation in 2016. In November 2017, it completed phase III clinical trials for treatment-resistant depression in the United States. Johnson & Johnson filed a Food and Drug Administration (FDA) New Drug Application (NDA) for approval on 4 September 2018; the application was endorsed by an FDA advisory panel on 12 February 2019, and on 5 March 2019, the FDA approved esketamine, in conjunction with an oral antidepressant, for the treatment of depression in adults. In August 2020, it was approved by the U.S. Food and Drug Administration (FDA) with the added indication for the short-term treatment of suicidal thoughts. Since the 1980s, closely associated ketamine has been used as a club drug also known as "Special K" for its trip-inducing side effects.
=== Pharmacodynamics === Similarly to MDMA, SDMA is a serotonin–norepinephrine–dopamine releasing agent (SNDRA) and a non-selective serotonin 5-HT2 receptor agonist. However, SDMA was 11-fold more potent as a serotonin releaser, 19-fold more potent as a dopamine releaser, and 2-fold more potent as a norepinephrine releaser than MDMA in HEK293 cells in vitro. In addition, it was about twice as potent as a serotonin 5-HT2A receptor agonist, whereas it showed similar agonistic potency as MDMA at the serotonin 5-HT2B and 5-HT2C receptors. SDMA had similar activational efficacies at the serotonin 5-HT2 receptors as MDMA. Due to its greater potency as a monoamine releasing agent, SDMA may be active at lower doses or concentrations than MDMA. SDMA produced hyperlocomotion and hyperthermia in rodents with similar profiles as MDMA. However, SDMA did not produce rewarding effects in the conditioned place preference (CPP) paradigm unlike MDMA. Hence, SDMA might have reduced misuse potential compared to MDMA. As with MDMA, SDMA did not produce the head-twitch response, a behavioral proxy of psychedelic effects, in rodents, and hence may not produce hallucinogenic effects in humans. SDMA might be less cardiotoxic than MDMA due to having much greater monoamine-releasing potency but unaltered serotonin 5-HT2B receptor agonistic potency.
Some individuals with CAIS may choose to retain their gonads. If this is the case, annual imaging of the gonads via MRI or ultrasound is recommended in order to monitor for signs of malignancy. Diagnostic laparoscopy and biopsy are also to be considered if imaging is ambiguous. A study in 2012 claimed that adult women with CAIS are increasingly likely to keep their gonads due to perceived benefits. A retrospective study which includes postpubertal CAIS individuals seen at a single UK DSD center shows that there has been a temporal shift away from gonadectomy. 76.5% CAIS individuals between 2020 and 2025 elected to retain their gonads in this study. Endogenous hormone profiles show very specific features that influence bone health, hormonal replacement therapy may improve bone mineral density, but it does not normalize it. For individuals with CAIS who wish to keep their gonads, a biannual screening program is proposed. It may be possible to avoid gonadectomy throughout the patient's life with regular imaging and routine monitoring to ensure the gonads are healthy. It is emphasized that not all imaging abnormalities are indicative of malignancy. Some activists consider that cancer risk is used to legitimate surgical and hormonal intervention because intersex bodies were positioned as inherently sick and in need of modification. Research also suggest that timely intervention to reduce genotoxicity such as DNA damage, inflammation and imbalanced autophagy may promote germ cells specification and decrease the risk of germ cells tumor.
Juxtarestiform body A bundle of fibers within the inferior cerebellar peduncle that connects the vestibular nuclei to the cerebellum, involved in balance and eye movement coordination. Junctional complex A structural region where neurons, glia, or epithelial cells make close contact. In the nervous system, includes tight junctions and adherens junctions that help form the blood–brain barrier.

==== Petitions to reconstitute former states ==== With the Paris Agreements in 1954, West Germany regained (limited) sovereignty. This triggered the start of the one-year period as set in paragraph 2 of Article 29. As a consequence, eight petitions for referendums were launched, six of which were successful:
=== Differential Diagnosis === While Kwashiorkor is primarily characterized by edema, growth failure, and dermatitis, it can sometimes be misdiagnosed as Acrodermatitis Enteropathica (AE), as both conditions present with similar dermatological symptoms. However, AE is caused by zinc deficiency, not protein deficiency, and is associated with a significant decrease in serum zinc levels. Unlike kwashiorkor, which is diagnosed based on clinical signs like edema and low serum albumin levels, AE requires additional diagnostic tests, such as zinc uptake tests and genetic screening, to differentiate it from other disorders with similar symptoms. Therefore, while both conditions can present with dermatitis, the underlying etiology helps distinguish the two.
Ribosomes are minute particles consisting of RNA and associated proteins that function to synthesize proteins. Proteins are needed for many cellular functions, such as repairing damage or directing chemical processes. Ribosomes can be either free, floating within the cytoplasm or attached to the rough endoplasmic reticulum. Their main function is to convert genetic code into an amino acid sequence and to build protein polymers from amino acid monomers. Ribosomes act as catalysts in two extremely important biological processes called peptidyl transfer and peptidyl hydrolysis. In summary, ribosomes have two main functions: Decoding the message, and the formation of peptide bonds. These two functions reside in the ribosomal subunits. Each subunit is made of one or more rRNAs and many r-proteins. The small subunit (30S in bacteria and archaea, 40S in eukaryotes) has the decoding function, whereas the large subunit (50S in bacteria and archaea, 60S in eukaryotes) catalyzes the formation of peptide bonds, referred to as the peptidyl-transferase activity. The bacterial (and archaeal) small subunit contains the 16S rRNA and 21 r-proteins (Escherichia coli), whereas the eukaryotic small subunit contains the 18S rRNA and 32 r-proteins (Saccharomyces cerevisiae, although the numbers vary between species). The bacterial large subunit contains the 5S and 23S rRNAs and 34 r-proteins (E. coli), with the eukaryotic large subunit containing the 5S, 5.8S, and 25S/28S rRNAs and 46 r-proteins (S. cerevisiae; again, the exact numbers vary between species).
The word "GULAG" is an acronym for Glavnoe Upravlenie Lagerei, or Main Camp Administration, the institution which ran the Soviet camps. But over time, the word has also come to signify the system of Soviet slave labor itself, in all its forms and varieties: labor camps, punishment camps, criminal and political camps, women's camps, children's camps, transit camps. Even more broadly, "Gulag" has come to mean the Soviet repressive system itself, the set of procedures that Alexander Solzhenitsyn once called "our meat grinder": the arrests, the interrogations, the transport in unheated cattle cars, the forced labor, the destruction of families, the years spent in exile, the early and unnecessary deaths.

Long-term storage is usually at minus 20 to minus 80 degrees Celsius in a sealed, desiccated container. Working aliquots can be held briefly at 2 to 8 degrees Celsius.
Repeated temperature cycling can promote aggregation and peptide degradation. Dividing material into single-use aliquots limits this risk.
Mass spectrometry is commonly used to confirm molecular mass and detect structural modifications. It is often paired with chromatographic purity assessment.
