Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.

In the spring of 1921, Banting traveled to Toronto to explain his idea to John Macleod, Professor of Physiology at the University of Toronto. Macleod was initially skeptical, since Banting had no background in research and was not familiar with the latest literature, but he agreed to provide lab space for Banting to test out his ideas. Macleod also arranged for two undergraduates to be Banting's lab assistants that summer, but Banting required only one lab assistant. Charles Best and Clark Noble flipped a coin; Best won the coin toss and took the first shift. This proved unfortunate for Noble, as Banting kept Best for the entire summer and eventually shared half his Nobel Prize money and credit for the discovery with Best. On 30 July 1921, Banting and Best successfully isolated an extract ("isletin") from the islets of a duct-tied dog and injected it into a diabetic dog, finding that the extract reduced its blood sugar by 40% in 1 hour. Banting and Best presented their results to Macleod on his return to Toronto in the fall of 1921, but Macleod pointed out flaws with the experimental design, and suggested the experiments be repeated with more dogs and better equipment. He moved Banting and Best into a better laboratory and began paying Banting a salary from his research grants. Several weeks later, the second round of experiments was also a success, and Macleod helped publish their results privately in Toronto that November.
In ancient times, one highly abrasive depilatory paste consisted of an admixture of slaked lime, water, wood-ash and yellow orpiment (arsenic trisulfide); In rural India and Iran, where this mixture is called vajibt, it is still commonly used to remove pubic hair. In other cultures, oil extracted from unripe olives (which had not reached one-third of their natural stage of ripeness) was used to remove body hair. During the medieval period, Catholic women were expected to let their hair grow long as a display of femininity, whilst keeping the hair concealed by wearing a wimple headdress in public places. The face was the only area where hair growth was considered unsightly; 14th-century ladies would also pick off hair from their foreheads to recede the hairline and give their face a more oval form. From the mid-16th century, it is said when Queen Elizabeth I came to power, she made eyebrow removal fashionable. By the 18th century, body hair removal was still considered a non-necessity by European and American women. But in 1760, when the first safety straight razor appeared for men to safely shave their beard and not inadvertently cut themselves, some women allegedly used this safety razor too. It was invented in Paris by the French master cutler Jean-Jacques Perret, author of La pogonotomie, ou L'art d'apprendre à se raser soi-même (Pogonotomy, or The Art of Learning to Shave). It was not until the late 19th century that women in Europe and America started to make hair removal a component of their personal care regime.
On sharp reverse curves, the buffers can become locked by slipping over, and on to, the back of each other. Although careful track design makes this a rare occurrence, a series of derailments at Stuttgart Central Station in 2012 were caused by buffer-locked wagons. Buffer-lock could occur on very sharp switches on rolling stock with the older, rounded buffers. Newer buffers are rectangular and are wider than they are tall. Buffers and chain couplers allow curves to have around 150 m (492 ft) radius, but sharp S-curves are not allowed. If it were not for the couplers, much sharper curves could be allowed, provided the train is not too long.
Several claims made by Hoyle and Wickramasinghe between 1977 and 1981, such as a report of having detected interstellar cellulose, were criticised by one author as pseudoscience. Phil Plait has described Wickramasinghe as a "fringe scientist" who "jumps on everything, with little or no evidence, and says it's from outer space".

=== Anne Marie West === In September 1972, the Wests led eight-year-old Anne Marie to the cellar at 25 Cromwell Street. The child was ordered to undress, with Rose tearing her dress from her body upon noting the child's hesitation. She was then stripped naked, bound to a mattress and gagged before Fred raped her with Rose's active encouragement. After the rape, Rose followed Anna Marie to the bathroom and handed her a sanitary towel, explaining to the child, "Everybody does it to every girl. It's a father's job. Don't worry, and don't say anything to anybody." Making clear these sexual assaults would continue, Fred and Rose then threatened the child with severe beatings if they ever received word she had divulged the sexual abuse she endured at their hands. Rose occasionally sexually abused Anne Marie herself, and later took extreme gratification in degrading her with acts such as binding her to various items of furniture before encouraging Fred to rape her, or forcing her to perform household chores while wearing sexual devices and a miniskirt. Fred and Rose forced Anne Marie into prostitution from the age of 13, telling clients she was aged 16. Rose was always present in the room when these acts occurred to ensure the girl did not reveal her true age. On one occasion when Anne Marie was aged 13 or 14, Rose took her to a local pub, insisting she drink several glasses of barley wine. Several hours later, Fred arrived at the pub to collect Rose and Anne Marie.
=== Polished === Mirror polished stainless-steel belts to produce several types of thin film and ceramic sheets. The belts are available in thickness from 0.60 to 2.00 mm (0.02362 to 0.0787 in.) with thickness variation less than or equal to 80 μm. Polished steel belts are produced endlessly or as open-length belts.

== Applications == Surfactants are widely used due to their ability to modify surface and interfacial properties, making them relevant in processes involving the interaction of hydrophobic and hydrophilic substances. Their amphiphilic nature—containing both hydrophilic and hydrophobic parts—enables them to bridge these otherwise immiscible components, thereby facilitating mixing and enhancing the efficiency of various physical and chemical transformations. This makes surfactants useful in numerous fields where control over interfacial interactions is relevant. Surfactants play an important role as cleaning, wetting, dispersing, emulsifying, foaming and anti-foaming agents in many practical applications and products, including detergents, fabric softeners, motor oils, emulsions, soaps, paints, adhesives, inks, anti-fogs, ski waxes, snowboard wax, in flotation, washing and enzymatic processes, and laxatives.
For even higher stresses, materials exhibit plastic behavior, that is, they deform irreversibly and do not return to their original shape after stress is no longer applied. For rubber-like materials such as elastomers, the slope of the stress–strain curve increases with stress, meaning that rubbers progressively become more difficult to stretch, while for most metals, the gradient decreases at very high stresses, meaning that they progressively become easier to stretch. Elasticity is not exhibited only by solids; non-Newtonian fluids, such as viscoelastic fluids, will also exhibit elasticity in certain conditions quantified by the Deborah number. In response to a small, rapidly applied and removed strain, these fluids may deform and then return to their original shape. Under larger strains, or strains applied for longer periods of time, these fluids may start to flow like a viscous liquid. Because the elasticity of a material is described in terms of a stress–strain relation, it is essential that the terms stress and strain be defined without ambiguity. Typically, two types of relation are considered. The first type deals with materials that are elastic only for small strains. The second deals with materials that are not limited to small strains. Clearly, the second type of relation is more general in the sense that it must include the first type as a special case.
== Measurement and values == Measurement is done from a sample of venous blood using immunological measuring mechanisms like ELISA, RIA, etc. Often these are done by automated machines to minimize human error.

Long-term storage is usually at minus 20 to minus 80 degrees Celsius in a sealed, desiccated container. Working aliquots can be held briefly at 2 to 8 degrees Celsius.
Repeated temperature cycling can promote aggregation and peptide degradation. Dividing material into single-use aliquots limits this risk.
Mass spectrometry is commonly used to confirm molecular mass and detect structural modifications. It is often paired with chromatographic purity assessment.
It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
