== Use == Chipotles impart a relatively mild but earthy spiciness to many dishes in Mexican cuisine. The chilis are used to make various salsas. Chipotle can be ground and combined with other spices to make a meat marinade – adobo. Chipotle is used, typically in powdered form, as an ingredient in homemade and commercial products, including some brands of barbecue sauce and hot sauce, as well as in some chili con carnes and stews. Usually, when used commercially, the product is advertised as having chipotle in it. Chipotles are spicy and have a distinctive smoky flavor. The flesh is thick, so the chilis are usually used in a slow-cooked dish rather than raw. They can also be lightly toasted on a dry comal or skillet until they are fragrant and slightly swell. When overcooked, they can be bitter. For some traditional Mexican sauces, the toasted chilis would be sautéed in oil or lard before being pureed. The chilis can also be soaked in warm water or stock until they become pliable and then can be added to a dish. == Biophysical and theoretical methods == Surface plasmon resonance (SPR) is the most common label-free technique for the measurement of biomolecular interactions. SPR instruments measure the change in the refractive index of light reflected from a metal surface (the "biosensor"). Binding of biomolecules to the other side of this surface leads to a change in the refractive index which is proportional to the mass added to the sensor surface. In a typical application, one binding partner (the "ligand", often a protein) is immobilized on the biosensor and a solution with potential binding partners (the "analyte") is channelled over this surface. The build-up of analyte over time allows to quantify on rates (kon), off rates (koff), dissociation constants (Kd) and, in some applications, active concentrations of the analyte. Several different vendors offer SPR-based devices. Best known are Biacore instruments which were the first commercially available. Dual polarisation interferometry (DPI) can be used to measure protein–protein interactions. DPI provides real-time, high-resolution measurements of molecular size, density and mass. While tagging is not necessary, one of the protein species must be immobilized on the surface of a waveguide. === AMDS === Research overview: In this study there are several atomic-scale molecular dynamics simulations created to illustrate how ethanol affects biomembranes containing phospholipids. The phospholipid membrane systems are comparable to the model membranes above but it only consists of one phospholipid which is palmitoyl-oleoyl-phosphatidylcholine (POPC) or palmitoyl-oleoyl-phosphatidylethanolamine (POPE). The primary difference between the phosphatidylcholine (PC) and phosphatidylethanolamine (PE) is that the three methyl groups attached to the Nitrogen atom for the PC structure is replaced by three hydrogen groups. The overall purpose of this study is similar to the study described above determining the effects of ethanol on biomembranes and how it is able to increase disorder in the membrane interior region forming non-lamellar phases in phospholipids. The experimental method and analytical technique is quite different. In the previous study, it emphasized the NNR technique using a set of host phospholipids, exchanging lipids, ethanol, and cholesterol to create model membranes. An aqueous solution containing 5% ethanol (v/v) was maintained but the concentration of cholesterol was varied to prove how this sterol compound can inhibit the effects of ethanol (inducing a liquid-disorder phase or non-lamellar phases) which is depicted in the different plots of the equilibrium constant (K) versus the mol% of cholesterol for each model membrane.
== Career and research == In 1961, he took a post-doctoral fellowship at the California Institute of Technology, working with George S. Hammond, who was an organic photo-chemist. Together, they found that some catalyzed reactions can occur up to one-million times faster than non-catalyzed reactions. Intrigued by this discovery, Knowles became an enzymologist. For a brief time, Knowles was a visiting professor at Yale University. in 1974, Knowles moved his research group to Harvard and became a professor there. Knowles's research was on the boundary of chemistry and biochemistry, and concerned the rate and specificity of enzyme catalysis and the evolution of enzyme function. Early in his career, Knowles studied α-chymotrypsin and pepsin, which are nonspecific proteases, meaning they accept a broad range of substrates.
