A real-time polymerase chain reaction (real-time PCR, or qPCR when used quantitatively) is a laboratory technique of molecular biology based on the polymerase chain reaction (PCR). It monitors the amplification of a targeted DNA molecule during the PCR (i.e., in real time), not at its end, as in conventional PCR. Real-time PCR can be used quantitatively and semi-quantitatively (i.e., above/below a certain amount of DNA molecules). Two common methods for the detection of PCR products in real-time PCR are (1) non-specific fluorescent dyes that intercalate with any double-stranded DNA and (2) sequence-specific DNA probes consisting of oligonucleotides that are labelled with a fluorescent reporter, which permits detection only after hybridization of the probe with its complementary sequence. The Minimum Information for Publication of Quantitative Real-Time PCR Experiments (MIQE) guidelines, written by professors Stephen Bustin, Mikael Kubista, Michael Pfaffl and colleagues propose that the abbreviation qPCR be used for quantitative real-time PCR and that RT-qPCR be used for reverse transcription–qPCR. The convoy scattered, and Gaddafi and those closest to him fled to a nearby villa, which was shelled by rebel militia from Misrata. Fleeing to a construction site, Gaddafi and his inner cohort hid inside drainage pipes while his bodyguards battled the rebels; in the conflict, Gaddafi suffered head injuries from a grenade blast while Jabr was killed. The Misrata militia took Gaddafi prisoner, causing serious injuries as they tried to apprehend him; the events were filmed on a mobile phone. A video appears to picture Gaddafi being poked or stabbed in the anus "with some kind of stick or knife" or possibly a bayonet. Pulled onto the front of a pick-up truck, he fell off as it drove away. His semi-naked body was then placed into an ambulance and taken to Misrata; upon arrival, he was found to be dead. Official NTC accounts claimed that Gaddafi was caught in a crossfire and died from bullet wounds. Other eye-witness accounts claimed that rebels had fatally shot Gaddafi in the stomach. One approach is to determine the optimal host system for each specific target protein product, as different, especially non-native proteins often have deviant behavior in other organisms, and some host systems may produce higher yields, or require more mild conditions than others. == Research == From the beginning of his career Bengt Mannervik studied enzymes of glutathione metabolism, including studies of levels in different tissues, structure and catalytic activity of glutathione transferase, a purification method, a detailed review on the isoenzymes of glutathione transferase, and many others. These publications have had a major influence on the field of glutathione biochemistry. Each of those mentioned above, together with three others, had been cited more than 1000 times by the end of 2024, the first more than 5000 times, leading to an h index of 89, as calculated by Google Scholar. In all he has had nearly 600 publications, with a combined total of more than 47000 citations. His interest in glutathione transferases has continued after his retirement, for example studies of their role as efficient ketosteroid isomerases and as enzymes involved in the biosynthesis of moulting hormones in mosquitoes transmitting malaria and yellow fever. In addition to the work directed specifically at enzymes involved in glutathione metabolism and detoxication, Mannervik coauthored texts on molecular toxicology. He also studied various more general aspects of enzymology, including graphical analysis, error structure of kinetic experiments, weighting of observations, regression methods, directed enzyme evolution, and discrimination between models. Major contributions were more recently directed to the evolution of novel functions by in vitro protein evolution.

== Limitations == In order to get a meaningful sulfur signal from the analysis, the buffer should not contain sulfur (i.e. no BES, DDT, HEPES, MES, MOPSO or PIPES compounds). Excessive amounts of chlorine in the buffer should also be avoided, since this will overlap with the sulfur peak; KBr and NaBr are suitable alternatives. Due to the low penetration depth of protons and heavy charged particles, PIXE is limited to analyzing the top micrometre of a given sample.
The mean improvement in scores with dextromethorphan/bupropion was statistically significant but not clinically significant relative to placebo at all assessed timepoints including at the end of week 1, although at the end of the study some patients did have clinically significant improvement. In Study 2 (STRIDE-1), dextromethorphan/bupropion was compared with bupropion alone in another randomized controlled trial. The dose of bupropion in the study was lower than the target dose recommended for clinical practice. In this study, dextromethorphan/bupropion showed significantly greater improvement than bupropion alone in the first two weeks of treatment but not by week 6 of treatment in people with major depressive disorder. The baseline scores were 33.4 points with dextromethorphan/placebo and 33.2 points with placebo, while the score reductions at week 1 were 5.2 points on the MADRS with dextromethorphan/bupropion and 3.6 points with bupropion (a 1.6-point difference), at week 2 were 8.0 points with dextromethorphan/bupropion and 6.1 points with bupropion (a 1.9-point difference), and at week 6 were 11.6 points with dextromethorphan/bupropion and 9.4 points with bupropion (a 2.2-point difference). On the basis of this trial, the FDA concluded that dextromethorphan contributes to the apparent antidepressant effects of dextromethorphan/bupropion.
