Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.

Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The aromatic acids 3-phenylpropionic acid (3PPA) and 3-(4-hydroxyphenyl)propionic acid (HPPA) are important commodities used in the chemical, pharmaceutical, and food industrial sectors. A biosynthetic route to synthetize these compounds was designed by combining the Escherichia coli phenylalanine pathway with non-native enzymes like tyrosine ammonia lyase (TAL) and the clostridial EnoR. The full pathway was assembled in Escherichia coli, which led to the production of HPPA and cinnamyl alcohol. Optimization of individual enzyme expression levels produced 3PPA and HPPA with a concentration of 367 and 225 mg/L, in this case the oxygen-sensitive EnoRs were catalytically active under the microaerophilic fermentation conditions. The 2-methylsuccinic acid (2-MSA) is used to synthetize polymers that have application as bioplastics, cosmetic solvents and coatings. A pathway for its synthesis was designed using Escherichia coli, combining native pyruvate and acetyl-CoA biosynthesis with methanogenic citramalate synthase (CimA), isopropylmalate isomerase (LeuCD), and the ER YqjM or KpnER from Klebsiella pneumoniae. The successful production of 2-MSA was achieved with a concentration of 0.96 g/L using KpnER.
=== Medical use === Urea-containing creams are used as topical dermatological products to promote rehydration of the skin. Urea 40% is indicated for psoriasis, xerosis, onychomycosis, ichthyosis, eczema, keratosis, keratoderma, corns, and calluses. If covered by an occlusive dressing, 40% urea preparations may also be used for nonsurgical debridement of nails. Urea 40% "dissolves the intercellular matrix" of the nail plate. Only diseased or dystrophic nails are removed, as there is no effect on healthy portions of the nail. Urea has been studied as a diuretic. It was first used by Dr. W. Friedrich in 1892. In a 2010 study of ICU patients, urea was used to treat euvolemic hyponatremia and was found safe, inexpensive, and simple. Like saline, urea has been injected into the uterus to induce abortion, although this method is no longer in widespread use. The blood urea nitrogen (BUN) test is a measure of the amount of nitrogen in the blood that comes from urea. It is used as a marker of renal function, though it is inferior to other markers such as creatinine because blood urea levels are influenced by other factors such as diet, dehydration, and liver function. Urea has also been studied as an excipient in drug-coated balloon (DCB) coating formulations to enhance local drug delivery to stenotic blood vessels. Urea, when used as an excipient in small doses (~3 μg/mm2) to coat DCB surface was found to form crystals that increase drug transfer without adverse toxic effects on vascular endothelial cells.
Top-down proteomics is a method of protein identification capable of identifying and quantitating unique proteoforms through the analysis of intact proteins. The name is derived from the similar approach to DNA sequencing. During mass spectrometry, intact proteoforms are typically ionized by electrospray ionization and analysed using a variety of mass analysers, including Orbitraps, Ion Cyclotrons and Time-Of-Flight. Effective fractionation is critical for sample handling before mass-spectrometry-based proteomics. Typical proteome analysis routinely involves digesting intact proteins followed by inferred protein identification using mass spectrometry (MS; Bottom Up proteomics). Top-down proteomics using mass spectrometry interrogates protein structure through measurement of a proteoform's intact mass followed by direct ion dissociation in the gas phase. Top Down proteoform analysis can also be achieved through resolution (separation) of the proteoform from all other proteoforms and then applying peptide-centric LC-MS/MS to characterise the isolated proteoform. A single gene can be coded for many protein products (e.g. via alternative splicing; post-transcriptional and -translational processing) and the resulting canonical amino acid sequences (i.e. 'proteins' or more correctly Open Reading Frame (ORF) products) can be further modified by any number of post-translational modifications (PTM) or non-physiological adducts. These varied protein species or proteoforms define proteomes and are the functional entities underlying biological processes.
Maduro's supporters protested against his capture in Caracas, with protesters holding portraits of Maduro and pro-Maduro and pro-Partido Socialista Unido de Venezuela (PSUV) banners. PSUV called for citizens to demand the release of Maduro and Flores. CNN reported that while some Venezuelans celebrated Maduro's ousting, others were skeptical of Trump's motives and condemned the capture of Maduro as a "kidnapping". In the United States, hundreds of people gathered in New York City's Times Square to protest against the operation, with people chanting "Hands off Venezuela" and holding up signs that stated "US Out" and "No Blood for Oil". Outside of New York, hundreds of people in different cities gathered to voice opposition to the strikes. One month after the strikes, the Venezuelan government called for a protest that gathered thousands of supporters for the release of Nicolás Maduro. His son Nicolás Maduro Guerra addressed the crowd, saying that the 3 January "will remain marked like a scar on our face, forever."
=== Nanometer-size clusters === Monodisperse, nanometer-size clusters (also known as nanoclusters) are synthetically grown crystals whose size and structure influence their properties through the effects of quantum confinement. One method of growing these crystals is through inverse micellar cages in non-aqueous solvents. Research conducted on the optical properties of MoS2 nanoclusters compared them to their bulk crystal counterparts and analyzed their absorbance spectra. The analysis reveals that size dependence of the absorbance spectrum by bulk crystals is continuous, whereas the absorbance spectrum of nanoclusters takes on discrete energy levels. This indicates a shift from solid-like to molecular-like behavior which occurs at a reported cluster the size of 4.5 – 3.0 nm. Interest in the magnetic properties of nanoclusters exists due to their potential use in magnetic recording, magnetic fluids, permanent magnets, and catalysis. Analysis of Fe clusters shows behavior consistent with ferromagnetic or superparamagnetic behavior due to strong magnetic interactions within clusters. Dielectric properties of nanoclusters are also a subject of interest due to their possible applications in catalysis, photocatalysis, micro capacitors, microelectronics, and nonlinear optics.

==== Aortic disease ==== A 2014 Cochrane review investigated the use of beta blockers in the maintenance of chronic type B thoracic aortic aneurysm in comparison to other anti hypertensive medications. The review found no suitable evidence to support the current guidelines recommending its use. A 2017 Cochrane review on the use of beta blockers to prevent aortic dissections in people with Marfan syndrome was unable to draw definitive conclusions due to lack of evidence.
A phase I clinical research study to test the safety and efficacy of a combination chemotherapy regimen consisting of methotrexate, L-asparaginase, idarubicin, and dexamethasone followed by allogenic or autologous bone marrow transplantation in 26 participants newly diagnosed with BPDCN is planned but not yet in its recruiting phase. While few studies have reported on the treatment of BPDCN that has recurred following initial therapy, donor lymphocyte infusions coupled with alternative chemotherapy treatments have induced second complete or partial remissions in a few patients.
=== Army leadership resignations === In April 2026, Hegseth fired Army Chief of Staff Gen. Randy George which has left the longest break without a service chief in Army history and making him one of eight high ranking members of the military fired or forced out by the secretary of defense. On August 31, 2026 Secretary of the Army Dan Driscoll gave Hegseth his resignation after a face to face meeting Driscoll had with President Trump where he stated his concerns over the number of army generals fired under Hegseth.
The defective protein can be transmitted by contaminated harvested human brain products, corneal grafts, dural grafts, or electrode implants and pituitary human growth hormone, which has been replaced by recombinant human growth hormone that poses no such risk. It can be familial (fCJD) or it may appear without clear risk factors (sporadic form: sCJD). In the familial form, a mutation has occurred in the gene for PrP, PRNP, in that family. All types of CJD are transmissible irrespective of how they occur in the person. It is thought that humans can contract the variant form of the disease by eating food from animals infected with bovine spongiform encephalopathy (BSE), the bovine form of TSE, also known as mad cow disease. However, it can also cause sCJD in some cases. Cannibalism has also been implicated as a transmission mechanism for abnormal prions, causing the disease known as kuru, once found primarily among women and children of the Fore people in Papua New Guinea, who previously engaged in funerary cannibalism. While the men of the tribe ate the muscle tissue of the deceased, women and children consumed other parts, such as the brain, and were more likely than men to contract kuru from infected tissue. Prions, the infectious agent of CJD, may not be inactivated using routine surgical instrument sterilization procedures.

== Medical uses == Selenium disulfide is sold as an antifungal agent in shampoos (such as Selsun Blue) for the treatment of dandruff and seborrheic dermatitis associated in the scalp with fungi of genus Malassezia. It is also used on the body to treat tinea versicolor (pityriasis versicolor), a type of fungal skin infection caused by a different species of Malassezia. A 2015 systematic review of topical treatments for seborrheic dermatitis of the scalp identified only a single randomized controlled trial evaluating selenium disulfide for the condition. It was a three-arm trial of 246 people with moderate to severe dandruff and compared treatment with 2% ketoconazole shampoo (n=97), 2.5% selenium disulfide shampoo (n=100), and placebo (shampoo base with no antiseborrheic agent) (n=49) for 29 days. The study found a 73% reduction in dandruff score with ketoconazole, a 67% reduction with selenium disulfide, and a 45% reduction with placebo. Based on the study, the systematic review concluded that selenium disulfide may be effective in the treatment of dandruff but that the available evidence is limited and overall evidence quality is low. It also found that while selenium disulfide has infrequent side effects, it seems to have more side effects than ketoconazole shampoo. Consequently, the review concluded that selenium disulfide should not be considered as a first-line therapy but instead should be used as an alternative treatment after other therapies like ketoconazole shampoo have proven not effective.
=== Statistics === In 2022, the sector with the highest number of companies registered in Denmark is Finance, Insurance, and Real Estate with 204,853 companies followed by Services and Retail Trade with 204,050 and 30,563 companies respectively.
=== Realization === The centrifugal partition chromatograph instrument is constituted with a unique rotor which contains the column. This rotor rotates on its central axis (while HSCCC column rotates on its planetary axis and simultaneously rotates eccentrically about another solar axis). With less vibrations and noise, the CPC offers a typical rotation speed range from 500 to 2000 rpm. Contrary to hydrodynamic CCC, the rotation speed is not directly proportional to the retention volume ratio of the stationary phase. Like DCCC, CPC can be operated in either descending or ascending mode, where the direction is relative to the force generated by the rotor rather than gravity. A redesigned CPC column with larger chambers and channels has been named centrifugal partition extraction (CPE). In the CPE design, faster flow rates and increased column loading can be achieved.
=== The Noffel clan === The Noffel clan was a criminal organisation that was founded by Naoufal "Noffel" Fassih. Naoufal moved to his cousin Abdelhadi Yaqout in Spain, at that time already one of the richest personalities in Marbella. Naoufal allied himself with the Taghi organisation, the Irish Mob and some members of the Martha organisation, to assassinate the billionaire drug lord Samir Bouyakhrichan in 2014. Naoufal was eventually spending more time in Ireland than in Spain. His relationship with Ireland's Kinahan clan was so strong that they even shared the same hideouts. It was also in an apartment of the Irish Kinahan clan where Naoufal was eventually arrested by the special forces of Dublin. He was sentenced by the Dutch court to life of imprisonment. Naoufal's cousin, Abdelhadi Yaqout, was a multi-millionaire businessman. He originated from Khouribga, in the south of Casablanca (Morocco), where he had spent a part of his childhood. After he emigrated to Spain, he became a very successful entrepreneur in Marbella. He was the owner of several clubs and bars in Puerto Banús, including the Rotana Shisha Lounge, the Portside bar, and the Tibu nightclub, which were frequently visited by Arab princes, Russian businessmen and celebrities. Yaqout was also a friend of the Anglo-Egyptian billionaire Mohamed Al-Fayed. Yaqout laundered money for the Kinahan clan, and he was a partner of Daniel Kinahan. According to the Spanish authorities, they had very close relations.

Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
